Comparison of two methods for radioassay of vitamin B12 in serum.

چکیده
A reference radioassay for vitamin B12 in serum, in which human serum is used as binder and DEAEcellulose is used to separate free and bound B12, was compared (194 sera) with a packaged kit (“Phadebas B12 Test”), in which intrinsic factor is used as binder and poiysaccharide for separation. The correlation coefficient was 0.949. With the kit, added B12 was identifiable, proportional dilution characteristics were excellent, and serial reproducibility was good. The normal range for the reference assay was 200-1000 pg/mi; all B12-deficient patients had values of <165 pg/mi. In a separate study, 38 patients with documented (clinical and laboratory) B12 deficiency had mean serum values of <150 pg/mi by the reference assay, 250 pg/mi by the kit. For the kit, the normal range was 250-1100 pg/mi. All patients with serum values of <150 pg/mi by kit assay were B12 deficient; false positives were seen in the 150-250 pg/mI range.

Vitamin B12 radioassay with oyster toadfish (Opsanus tau) serum as binder.
We describe a vitamin B12 radioassay in which oyster toadfish (Opsanus tau) serum is used as the binding protein. The serum is quite stable and has a high capacity and high binding affinity (K greater than 10(12) liter/mol) for vitamin B12. The binding is not significantly affected by temperature, the presence of denatured proteins, or the amount of vitamin B12 present. The radioassay is reprod...
متن کاملA Radioassay for Serum B 12 Using Unsaturated Transcobalamin
I N THE PAST SEVERAL YEARS a number of radioassay procedures for serum vitamin B12 have been described. Each method is based on the fact that unlabeled crystalline B12 will competitively inhibit isotopically labeled B12 (tracer B12) from binding to a specific B12 binding protein(s). Basic to each procedure is the reaction of B12 (labeled and unlabeled) with a limited quantity of binding protein...

متن کاملTwo radioassays for serum vitamin B12 and folate determination compared in a reference interval study.
For 154 subjects, we verified that vitamin B12 and folate status was normal, using as criteria the average polymorphonuclear lobe count, mean corpuscular volume, and hemoglobin concentration. We then used blood from these subjects to compare values obtained with two radioassay kits, each designed for simultaneous vitamin B12 and folate determination. Although regression analysis showed reasonab...
